Solutions for Chromatography Exercises 1. A main cause of band broadening is diffusion of the component from a region of high concentration to a region of lower concentration. 2. Plate height (H) is a measure of how efficient a chromatography column is in separating components. It is defined as where σ is the standard deviation, in units of length along the column, of the analyte concentration profile in its band, and L is the length of the column. Plate height is calculate from the number of plates associated with the column by H = L / N. A smaller plate height indicates that there is less band broadening (smaller σ) as a component moves through the column and is more desirable than a larger plate height. 3. 4. Since Rs is slightly larger than 1.5 the peaks have baseline resolution and do not overlap significantly.
