(HEMATOLOGY-2) (Leukocytes, ESR) 1. MEASURING BLOOD SYMPTOM RATE (LED) - Westergreen Method(Erythrocyte Sedimentation Rate = ESR) (Blood Bezinking Znelheid= BBS) Principle The blood sample with anticoagulant is put into a special scale tube and placed upright, then the erythrocytes will settle.This deposition was measured at 1 hour and 2 hours later. Tools & Reagents 1. Westergreen Pipette -Long : 300mm Diameter : 2.5 mm -Scale : 0 to 200 Fill the tube : -1 ml 2. Westergreen Tube 3. Westergreen shelves 4. Anticoagulants -Na Citrate 3.8% : 0.2 ml for every 0.8 ml of blood -EDTA : 1 mg for every 1 ml of blood Need to be diluted with 0.9% NaCl (4 volumes of blood: 1 volume of 0.9% NaCl) Work Procedures 1) Pipette 0.9% NaCl with a Westergreen pipette to a scale of 150, then put it in a Westergreen tube. 2) Blood samples with EDTA anticoagulant were sucked with a Westergreen pipette containing 0.9% NaCl. 3) Mix the contents of the Westergreen tube by sucking and blowing several times until mixed well. 4) The solution mixture in the Westergreen tube is then sucked with a Westergreen pipette until the scale is 0, then place the Westergreen pipette perpendicular to the Westergreen rack. 5) Read the height of precipitation at 1 hour and 2 hours. Reference Value Hour I: 0 – 2 mm Hour II: 2 – 11 mm
