SEDIMENT EXAMINATION Principle Spin a small amount of urine at low speed, then examine the precipitate (sediment) that forms under a microscope. TOOLS & REAGENTS 1) Test tube 2) Glas objects 3) Cover glass 4) Microscope 5) Centrifuge (+ centrifuge tube) 6) Urine sample WORK PROCEDURES 1) Pour 8 ml of urine sample into a centrifuge tube 2) Spin on low speed for 5 minutes(so as not to damage certain formations) 3) Then discard the supernatant(decanthe until approximately 0.5 ml remains) 4) Shake again until homogeneous, take 1 drop and put it on the object glass, cover with a cover glass 5) Observe under a microscope in a horizontal positionObservations were made with weak light, - lower the condenser - the diaphragm is slightly closed D.INTERPRETATION - With a 10X objective - Check the entire field of view at a glance and then pay attention if any crystals and pistons are found. - Calculate the number of small visual fields (/lpk). - With a 45X objective - Pay attention and calculate the field of view of the other formations. - The number of pistons reported has an average of small visual field (/lpk). - The average number of erythrocytes and leukocytes is reported as large visual field (/lpb). - Sediment elements that are less significant can simply be reported with a sign - + , available - + + , Lots - + + + , Many E.NOTES (Name the formations that can be found in acidic urine and alkaline urine, and draw these formations!Which types of crystals easily form urinary tract stones?)
