Vibrio Test on Fishery Products B. Work Procedures Vibrio cholera test 1. Weigh the TCBS media according to the size to be used, make TCBS media without needing to be autoclaved but still aseptic and hygienic. Then dissolve it with a hotplate or water bath until completely dissolved, the media is ready to use 2. The enrichment stage is carried out by inoculating 10 grams of sample at 90 mL of APW and then pipette 1 mL of each sample into 9 mL of APW and incubated for 24 hours at 37oC . 3. The probe test is carried out by streaking one loop of the enrichment culture on TCBS agar and incubated at 37oC for 24 hours. V. cholerae colonies will yellow with a flat surface, the middle is cloudy and part the edges are clear. 4. The reinforcement test is carried out by taking two separate colonies that show typical V.cholerae colonies and inoculated on TSI agar slants and SIM agar like in the Salmonella-Shigella test. Incubation was carried out at 37o C for 24-48 hours, then the reaction formed is checked against the TSI and SIM tables. As a control, streak the V.cholerae culture onto TCBS agar and streak also onto TSI and SIM agar to see the reaction that occurs.
