Calculation of Leaf Surface Area, Estimated Evaporation Rate and Dorsiventral Surface Transpiration of Leaves Objective: Calculating the surface area of leaves and the rate of evaporation and transpiration from leaves Materials and tools: Plant material : Leaves of several types of plants Tools : Analytical Scales, straw paper, paper clips, tape, scissors, Vaseline Ways of working: a. Calculate the leaf area 1. Take leaves from the plant (3 sheets), then stick them on a piece of paper whose weight and area are known. 2. Next, trace the leaf onto the paper. 3. Then trace the image of the cut leaf and weigh it. 4. Thus the leaf area can be calculated using the formula:  The weight of the leaf cutouts  Leaf area = x area of paper  Paper weight b. Estimated Leaf Evaporation Rate 1. Take a leaf whose surface area is known, then weigh it and hang it with a paper clip indoors or in direct sunlight. 2. At certain time intervals (30 minutes) the leaves are weighed (weighing is done 3 times). 3. Make a calculation list of the reduction in leaf weight during evaporation. Two leaves of known size in experiment a are weighed, then soaked in water and dried with tissue paper. 2. The first leaf is smeared with vaseline on its upper surface and the second on its lower surface, and weighed again. 3. Place the two leaves in the hot sun for 1 hour or more, and weigh again. 4. Compare the results between cuticular transpiration from the upper surface and stomatal transpiration from the lower surface. Experiment b. Stomata Structure and Stomata Opening and Closing Activities Objective: Know the general structure of stomata and the process of opening and closing stomata Materials and Tools: Material : Leaves of various monocot and dicot plants Tools : Microscope, object glass, cover glass, 1M sucrose or NaCl solution. Ways of working: 1. Drop distilled water on the surface of the glass object 2. Make a thin incision on the surface of the upper and lower epidermis of a leaf of the specified type, then place it in a drop of distilled water on a glass object, determine the upper epidermis and lower epidermis. 3. Cover carefully with a cover glass and observe under a microscope with small magnification (4x10) 4. Focus your observations on 1-2 stomata and increase the magnification to 40x10, then describe the structure of the stomata observed under the microscope. 5. Drizzle one part with sucrose and on the other side suck up the distilled water using a tissue so that the distilled water is replaced by sucrose and observe the changes that occur in the stomata. Note the time required for the process to occur and observe it. 6. Then drip it again with distilled water on one side while sucking sucrose on the other side, observe the changes that occur and note the time required for these changes. Place the observation in direct light so that the stomata respond with distilled water. 7. Then drip with NaCl by sucking distilled water on the other side and observe the changes that occur in the stomata, note the time required for these changes 8. Drop again with distilled water to see the response of the stomata, observe the time required for these changes. Describe the processes that occur sequentially. 9.
