Classification of Immobilization Methods Whole cell immobilization methods are broadly classified into physical methods, chemical methods, and biological methods. 5. Physical Methods of Whole Cell Immobilization 5.1 Adsorption Adsorption is the simplest method of whole cell immobilization. In this method, microbial cells are attached to the surface of solid carriers through weak physical forces such as van der Waals forces, hydrogen bonds, and ionic interactions. Commonly used supports include activated charcoal, glass beads, silica gel, alumina, and clay. Advantages: Simple, inexpensive, no toxic chemicals required. Disadvantages: Weak binding, chances of cell leakage, not suitable for long-term use. Applications: Wastewater treatment, biosensors, immobilized bioreactors. 5.2 Entrapment In the entrapment method, microbial cells are physically trapped inside a porous gel or matrix. The pores of the matrix allow substrates and products to diffuse freely while retaining the cells inside. Common entrapment materials include calcium alginate, agar, carrageenan, and polyacrylamide. Alginate entrapment is the most widely used method. Advantages: High cell density, protection from mechanical stress, widely used in industries. Disadvantages: Diffusion limitation, gradual leakage of cells. Applications: Ethanol fermentation, organic acid production, antibiotic synthesis. 5.3 Encapsulation Encapsulation involves enclosing microbial cells within a semi-permeable membrane or capsule. The membrane allows nutrients and products to pass through but retains the cells inside. Materials such as alginate-polylysine, cellulose nitrate, and synthetic polymers are commonly used. Advantages: Better protection and controlled mass transfer. Disadvantages: Costly and complex method. Applications: Probiotics, medical biotechnology, bioremediation. 6. Chemical Methods of Whole Cell Immobilization 6.1 Covalent Binding In covalent binding, microbial cells are chemically attached to solid supports through covalent bonds. The carrier surface is first activated using cross-linking agents such as glutaraldehyde or carbodiimides, followed by attachment of cells. This method provides strong and stable immobilization. Advantages: Strong attachment and minimal cell leakage. Disadvantages: Toxic chemicals may reduce cell viability. Applications: Continuous bioreactors and biosensor development. 6.2 Cross-Linking In cross-linking, microbial cells are directly linked to each other using bifunctional reagents such as glutaraldehyde, without using a carrier material. This results in the formation of a dense cell mass. Advantages: No support required and high cell concentration. Disadvantages: Reduced metabolic activity due to chemical toxicity. Applications: Industrial biotransformation processes. 7. Biological Method: Biofilm Formation Biofilm formation is a natural method of immobilization in which microbial cells adhere to a surface and produce extracellular polymeric substances, forming a stable biofilm.
