Experiment preliminary test:- precipitation by Heat Take 10ml of a given solution ina test tube and heat the upper portion of the test tube in direct flame Iflame, by holding the test tube ing the Handing position. Add few drops of 1% Acetic ecid. Do not disturb the text tube. precipitation by curdic Reagent To 1 ml of the given solution add 1 ml of picric acid precipitation by acidic Reagent (organic Reagent). Solvent To 1ml of solution add an equal amount of ethanol. • Coloured Reaction:- Biuret text:- To 2ml of sample add 2ml of 5% Sodium Hydroxide and 1 drop of 1%. Copper sulphate solution, Ninkyahin text:- To 2ml of Sample, add 10 drops of ninhydrin reagent heat to boiling. xanthoproteic test- To 3ml of sample add 1 ml. of cone. Nitric acid. heat it for Imin ard cool undertap water. Note the yellow Colour in acid medium, Divide the contents into two parts. To one half add few drops of 40% Sodium Hydroxide. - Millon's Testi- (Cole's test) To 1 ml of sample add 1ml of mercuric Sulphate reagent. boil gently for 30 see add & drops of 1% Sodium nitrite - Sakaguchi Test To 3ml of sample adol 2 drops of 40%. NaoMadd 1 crops of a-naphthol. Add few drops. of bromine Water. - Sulpher Testir To 2ml of Sample, add equal volume of 40% NaOH and boil at least for I min. cool and add 2-3 drops of lead acetate and mix Molisch's Test (Test for glycoproteins):.... To 1 ml of sample adel 2-3 drops of-naphthol and 1 ml of Cone Sulphuric acid along the Bides of the test tube.
