Determining the Effects of Moisture Levels in Soil on Bacterial Diversity November 14, 2024 Research Question: How do moisture levels in soil affect bacterial diversity? Do bacteria rely on moisture levels in their environment to grow? Location: Soil samples will be collected 5 meters from previous sites, starting at the river shore, with all samples taken from shaded areas. Sample sites 1. Sample 1: From the shore of the river. 2. Sample 2: 5 m away from Sample 1. 3. Sample 3: 5 m away from Sample 2. Here, note that all of our samples were taken form shaded area Equipments:trowel, plastic zip-lock bag Independent Variable: Moisture level in soil Dependent Variable: Bacterial diversity(the number of different colonies formed) Hypothesis:The level of moisture within the soil will influence the Bacterial growth Prediction: Increased moisture in soil will result in increased bacterial colony formation Procedure Soil will be collected from 3 different locations with differing levels of proximity to water using a trowel. Before collecting the sample, the moisture level will be recorded using a moisture reader. The sample collected and sealed in zip-lock bag The microbes in the sample will be isolated and grown on ager plates containing antibiotics The different types of colonies will be counted to determine the bacterial diversity in each sample
