1. Slide Test Method Using 10% Cell Suspension Objective : To reconfirm the donor's blood type examinationbefore transfusing to the patient Tools and materials: a. Object Glass b. Mixer c. 10% donor erythrocyte cell suspension d. Anti-A serum is usually blue or green in color e. Anti-B serum is usually yellow f. Anti-AB serum is usually pink/colorless g. Anti-D (Rhesus) serum is usually colorless / clear Ways of working : 1. Drop 1 drop (±50 µl) of anti-A, anti-B, anti-AB, and anti-D on the glass object 2. Pipette 50 µl of 10% donor cell suspension onto a glass object that has been treated with antisera 3. Stir with a stirrer each mixture of donor blood with antisera and shake 4. Observe whether there is agglutination macroscopically Reading results: Agglutination : there are antigens on the donor's red blood cells No agglutination : there are no antigens on the donor's red blood cells (Example of reading blood group results using the slide test method) 2.. Centrifuge d. 5% donor erythrocyte cell suspension o Anti-A serum is usually blue or green. o Anti-B serum is usually yellow e. Anti-AB serum is usually pink/colorless f. Anti-D (Rhesus) serum is usually colorless / clear Ways of working 1. Pipette 50 µl of anti-A, anti-B, anti-AB, and anti-D into each tube 2. Pipette 50 µl of 5% donor erythrocyte cell suspension into a tube containing antisera and homogenize 3. Centrifuge at a speed of 1000 rpm for 60 seconds 4.
