CONCENT RED BLOOD CELL WASHING Principle : With the addition of saline solution and rotationit will produce red blood cells that are free of protein or globulin Objective : To obtain concentrated red blood cells that have beenWashed so that it is free from protein/globulin Utility : 1. Preparation for making red blood cell suspension 2. Preparation for determining blood group antigens Method : Tube test Material : Red blood cells Reagents : Saline 0.9 % Equipment : 1. Centrifuge 2.Spray Pumpkin 3. Reaction Tube 4. Pasteur pipette 5. Tube rack 6. Parafilm Ways of working : 1. Prepare a test tube 2. Label according to the sample identity 3. Pipette as many concentrated red blood cells as possible 4. Add 0.9% saline to ¾ of the tube 5. Shake well with a Pasteur pipette or cover the tube with parafilm and turn the tube upside down until evenly mixed. 6. Rotate the tube at 3000 rpm for 2 minutes 7. Using a pipette, remove the supernatant until the red blood cells become concentrated again (100%) (Up to this point, the red blood cells have been washed once) 8. Repeat point 4- point 7 for the next wash (as needed) 8 drops into the tube Screening Results: Concentrated Red Blood Cells that have been washed (100%)
