The Action of Buffer Systems Most of the buffer systems in the body consist of a weak acid and the salt of that weak acid. Different Buffer Systems are: Plasma Buffer System/Extracellular Buffer System Bicarbonate Buffer Phosphate Buffer Protein Buffer RBC Buffer/Intracellular Buffer System Hemoglobin Buffer Phosphate Buffer Bicarbonate Buffer Urine Buffer Ammonia Buffer Phosphate Buffer Bicarbonate Buffer 2. RBC buffer / Intracellular buffer system Hemoglobin buffer Phosphate buffer Bicarbonate buffer 3. Urine buffer Ammonia buffer Phosphate buffer Bicarbonate buffer The principal buffer system of the fluid is: Protein buffer system Carbonic acid-bicarbonate buffer system Phosphate buffer system When pH rises, the free carboxyl group of one end of a protein can act like an acid by releasing a H+ ion. This H+ ion is then buffered with excess OH- in the solution to form water. R NH2-C-COOH → NH2-C-COO- + H+ H The free amino group (NH2) of the other end of a protein can act as a base by combining with a H+ ion when pH falls. So proteins can buffer both acid and base. R NH2-C-COOH + H+ → NH3+-C-COOH H The protein hemoglobin (Hb) is an important buffer at low pH. As CO2 from tissues combines with Hb to form carbonic acid (H2CO3), it dissociates into H+ and HCO3-.
