1. 4 types of receptors: - ligand gated ion channels - G protein coupled receptors (GPCR) - kinase receptors - nuclear/intracellular receptors 2. Describe ligand-gated ion channels: - also known as ionotropic receptors - directly coupled to ion channels - examples of these receptors = nicotinic 3. Where are nicotinic receptors found? - plasma membrane of cells - occurs in the NMJ junction, autonomic ganglia + CNS 4. Describe what happens - the ion channel opens when ACh binds to nico- - the plasma membrane's Na+ + K+ permeability tinic receptor: so depolarisation of the cells occurs due to rapid influx of Na+ - intracellular side of the plasma membrane is negatively charged compared to the extracellular surface - therefore the inward rush of positively charged Na+ ions reduces the negative membrane potential to cause depolarisation 5. What is the function + to transmit AP across somatic nerves location of NMJ (neuromuscular junction)? 6. Where does ACh synthe- in motor neuron terminal sis occur? - acetyl groups are donated by acetylcoenzyme A which is covalently bonded to choline to form ACh - the ACh is then stored in vesicles ready for release - when APs conducted along motor nerve terminals, depolarisation causes an influx of Ca2+ ions 7. During AP's (ACh): - influx of Ca2+ ions is caused - ACh is released into the synaptic cleft - vesicles now fuse with the terminal membrane 1/2 - released ACh now diffuses across the synaptic cleft + binds to nicotinic receptors on the sarcolemma, causing a depolarisation which can stimulate an AP in the muscle fibre - ACh in the synaptic cleft also interacts with the enzyme acetylcholinesterase which hydrolyses it to choline + acetic acid 2/2
