EXPERIMENT II ANALYSIS OF AMINO ACID Amino acids as protein monomers are organic molecules low number of molecules (between 100-200) containing at least one carboxyl group (COOH) and one amine group (NH2). There are 20 types of constituent amino acids proteins. The variation between amino acids lies in the type of side chain (R group). Based on the chemical properties of this R group, the properties of an amino acid can be predicted, instead knowledge of the nature of amino acids will help in identification R group. Amino acids are classified into seven groups based on chemical properties R group, so it will be easier to remember the general properties of each amino acid. With this classification, specific analysis methods can be designed for certain amino acids. 1.Ninhydrin Test Test ninhydrin ninhydrin is test general For sour amino. If (triketohydrindene hydrate) is heated with amino acids then it will colored complexes are formed. In this reaction NH3and CO2released so possibility can be measured quantitatively. Proline and hydroxyproline produce a complex that differs in color from other amino acids. Color complex formed contains two molecules of ninhydrin which react with ammonia after amino acids are oxidized. The overall reaction of amino acids with ninhydrin is as following: 1. Oxidative decarboxylation of amino acids and production of reduced ninhydrin, ammonia, and carbon dioxide. 2. Reaction of reduced ninhydrin with other ninhydrin molecules and ammonia released. 3. A blue complex is formed. Reactor : - Ninhydrin solution - 2% Protein Sample - 2% Amino Acid Sample Procedure : - Add 1 drop of 0.1% ninhydrin solution inside Heat the mixture until it boils. Observe the color changes that occur. 3 mL sample solution.
