TOTAL PROTEIN EXAMINATION METHOD Photometric test based on the biuret method PRINCIPLE Together with copper ions, the protein forms a blue violet complex in alkaline solution. Color absorbance is directly proportional to concentration. TOOLS AND MATERIALS Tool: • Micropipette • Tissue • Aquadest • Test tube • Spectrophotometer • Beaker Glass • Spray bottle • Blue Tip and Yellow Tip • Micropipette Material: • Monoreagent(1:4) R1:R2 • Aquadest • Standard • Reagents mentioned:R1:Sodium hydroxide 100mmol/L Potassium sodium tartrate 17mmol/L R2:Sodium hydroxide 500mmol/L Potassium sodium tartrate 80mmol/L Potassium iodide 75mmol/L Copper sulfate 20mmol/L WAYS OF WORKING 1. Use PPE properly, correctly and completely 2. Prepare the tools and materials that will be used 3. Ensure that all tools and materials are ready to be used 4. Prepare three test tubes, each filled with labels with standards, blanks and samples 5. Pipette 500 µL of monoreagent each then put into each of the three test tubes 6. Pipette 10 µL of distilled water and put it in a blank tube 7. Pipette 10 µL of standard reagent and put it in a standard tube 8. Pipette 10 µL of serum sample and place it in a sample tube 9. Homogenize and incubate for 5 minutes, at a temperature of 20-25°C / 37°C and read the absorbance for no more than 60 minutes. 10. Read the absorbance of the sample at a wavelength of 546 nm 11. Interpret the results of the total protein examination obtained INTERPRETATION OF RESULTS Mature: 6.6-8.8 g/dl Children Woman Man 1-30 days 4.2 - 6.2 g/dl 4.1 - 6.3 g/dl 1-6 months 4.4 - 6.6 g/dl 4.7 - 6.7 g/dl 6 months-1 year 5.6 - 7.9 g/dl 5.5 - 7.0 g/dl 1-18 years 5.7 –8.0 g/dl 5.7 –8.0 g/dl
