● EDTA ○ Usually seen as purple color (sometimes pink) ○ Chelates calcium CA2+ prevents blood clotting ■ FBC ● Maintaining RBC and WBC, and platelets morphology ● Hematology tests ○ ESR, Blood films ● Used for preserving cellular structures ○ Not appropriate for Calcium or coagulation ■ False indication of calcium (low) ■ Prolonged duration of clotting times ■ False indication on magnesium and Potassium ● EDTA a chelating agent ● Does not binds to Calcium magnesium, iron, zinc ○ Platelet clumping when not mixed ○ Over filtering ■ Alteration on blood ○ EDTA is not selective ■ Grabs onto anything (divalent or trivalent cations) ■ Prevents clotting ● Removes Ca2+ = needed removal for clotting to start ● Factor IV for clotting cascade ○ Enzyme assy needs Mg2+ as cofactor ■ Alkaline phosphatase ■ DNA polymerases ● Sodium Citrates ○ Sodium salt of citric acid ○ Used in trisodium citrate ○ Reversible Calcium chelators ○ Binds to ionized calcium ■ Reversible ● calcium - citrate bonds are not permanents ● Test such as PT, APTT will analyze excess calcium chlorides ○ Holds calcium in place temporarily ■ Still can be pulled off if need more calcium. ● Lymphocytes ○ Agranulocytes = round, large, dark staining ○ Subtypes ■ T lymphocytes ● Mature in thymus ● Cell mediated immunity ■ Helper T cells ● Activated through macrophage ● B cells + cytotoxic T cells ○ Kills virus invected cells ■ Regulatory T cells ● Suppression of immune response ● Maintenance of tolerance ○ B-lymphocytes ■ Maturation in Bone marrow ■ Antibody productions ○ NK cells ■ Innate immunity ■ Kills virus infected cells ● Does not need prior sensitization
