AIM: To prepare a temporary stained slide of human blood ma Materials required: Needle or blood lancel, cotton, slides, spirit Leishman's stain, microscope etc... PROCEDURE: 1. Place Two clean dry slides on the table. sterilise one of your fingers as well as the needle with Sprit prick the tip of the finger with the needle, press the finger, the blood, quite coge oud. put a chop of blood on one end of the clean slide. Immediately keep another slide at an angle of 45 on the first slide touching the blood push the upper slide towards other end rapidly to get a uniform film of blood. Let it dry. pour a few drops of Leishman's stain on the blood film After 2 to 3 minutes, drain out the stain and dip the stid in water to crash Dry the slide and observe under microscope OBSERVATIONS: The Following 8 types of blood cells or corpuscles are observed 1. Erythrocytes ar Red Blood corpulses (RBC) They are biconcave, lis like, red in coloxer, without e nucleus and must numerous. 2. Leucocytes or white blood copusclus (HI.B.C) They are large, nucleated and less numerous. They are of Pollowing typer. a) Granulocytes: They have granular cytoplasm and label nucleus. They are B types, 1) Neutrophils: found in large numbers, nucleus is lobed 1-s loby converted by thin: strand granular cytoplasm ⅱ) Ecsinophily (Audophily): Granular cytoplasm, bilobed nucleus, tobes not central, but along the periphery. Basophils Granulaz ectoplasm, lobed nucleus 20şg blobs, lobes less distinctly separated b) Agranulocytes. They have a clear (without granule) cytoplasm and decked nucleus. De are of two types.
