B-cell Activation/Differentiation and Antibody Effector Function Review Clonal Selection/Expansion Hypothesis: Each B-cell bears a single type of Ig receptor. Upon stimulation, each cell will create a clone of cells bearing the same antigen receptor as the original. Stimulation occurs when it meets its cognate (corresponding cell) or its specific antigen. Some of the clones will remain after the original immune response is complete. The memory cells are capable of an enhanced immune response. → Stronger binding affinity Plasma cell: A B-cell that has encountered an antigen and is secreting antibody. Current developmental phase: mature but naive. Moving from one 2° lymphoid organ to the next. B → Z B-cell = B-cell 1. Dependent vs T-Independent B-cell Responses: T stands for thymus. T-cell dependent / T-cell independent CD4⁺ (Helper) T-cell A B-cell can be activated without a T-cell. T1: Thymus independent Antigen binds to BCR and TLR. T2: Antigen binds to BCR w/o co-stimulatory molecules. T3: Thymus dependent? Activated by binding with T-cell. T-1 Antigen Different binding ligand Will be activated, but not as strong → Classical response comes from T-dependent. Isotype switching, affinity maturation, immunological memory only with T-cell activation.
