LEUKOCYTE EXAMINATION Leukocytes (white blood cells) are clear and colorless, they are larger of red blood cells, but the number is smaller. In every cubic millimeter of blood there are 6000 to 10000 (average 8000) white blood cells. The function of white blood cells is as a soldier of the body, namely to kill and eat disease germs/bacteria enters the body's RES tissue (reticuloendothelial system), where it reproduces in the spleen and lymph nodes; and as a carrier, namely transporting/carrying substances : Fat from the intestinal wall through the spleen continues to the blood vessels. Implementation of Practicum a. Tools and materials- Leukocyte thoma pipette - Sterile lancet Improved Neubauer counting room Turk's reagent 70% alcohol cotton Microscope b. Work procedures 1. Blood is sucked with a leukocyte pipette to the 0.5 mark. Excess blood on pipette tip is cleaned. 2. Suction Turk's reagent to mark 11 on the pipette (20x dilution), then make homogeneous by shaking the pipette for 3 minutes. 3. The prepared counting chamber is filled with blood + Turk (Note how to fill the counting room) 4. Leave it for 3 minutes then look under a microscope with 40x magnification. 5. Calculation for leukocytes: Number of leukocytes (/µl blood) = Vb = 4 x L x W x T = 4 x 1 x 1 x 0.1 = 0.4 µL blood Fp = volume of leukocytes taken n = Number of leukocytes (white blood cells) counted in the counting room Fp = Dilution factor Vb = calculated plane volume Normal values: Women = Men (5000-10000 / µL blood)
